Carbon nanotubes have emerged as promising nanostimulants to enhance
germination and early development in various cultivated species, but their effect on plants with
CAM metabolism, particularly in the genus Agave, remains unexplored. A. salmiana Otto ex
Salm-Dyck is a species of great ecological, cultural, and economic relevance in Mexico, whose
seed propagation faces limitations associated with low germination and progressive deterioration
during storage. This study evaluated the effect of multi-walled carbon nanotubes (MWCNT) on
germination, early development, and greenhouse survival of A. salmiana, considering the
interaction with seed storage time. Four seed lots with different collection years (2015, 2016,
2022, and 2024) were used. In the first phase, seeds from the most recent lot (2024) were exposed
to seven MWCNT concentrations (0, 25, 50, 75, 100, 125, and 150 mg L⁻¹) for 24 and 48 h to
select representative doses. In a second phase, the selected concentrations (50, 125, and 150 mg
L⁻¹) were applied to all four lots for 24 h, evaluating germination, leaf and root length, viability
(tetrazolium test), seed morphometry, survival, and greenhouse growth. Results showed a clear
dose-dependent pattern: the 50 mg L⁻¹ concentration significantly increased germination (up to
88% in recent seeds) and stimulated seedling growth, particularly in root length. Higher
concentrations (125–150 mg L⁻¹) generated contrasting responses, promoting root development
in some lots but reducing greenhouse survival in others. Storage time proved critical: the 2015
lot showed nearly null germination and viability (<10%), while the 2022 and 2024 lots achieved
germination percentages close to 90% and greenhouse survival above 85%. Surprisingly, the
2024 lot, despite having the largest and heaviest seeds, showed low viability in the tetrazolium
test (14.7%), indicating that morphometry is not a reliable predictor of physiological quality.
Together, these results demonstrate that MWCNT can act as effective biostimulants in A.
salmiana when applied at moderate concentrations (50 mg L⁻¹) to seeds with adequate
physiological quality (<3 years of storage), but their effect is nullified or becomes inhibitory in
lots with advanced deterioration or at high doses. This study provides the first evidence of nano-
bio-interactions in agaves and establishes methodological bases for incorporating
nanotechnology into propagation and germplasm conservation programs for species with CAM metabolism.

Background: Agave potatorum, commonly called tobalá or papalomé, is used for mezcal production in different states
of the Mexican Republic, with Oaxaca being the main producer. This species depends exclusively on sexual
reproduction for its propagation, since it does not produce vegetative propagules, so its conservation and perpetuation
depend entirely on seeds, which have reported a low viability rate, placing this species in a state of vulnerability.
Objective: To evaluate the effect of multi-walled carbon nanotubes (MWCNT) and gibberellic acid (AG₃) on
germination, initial development and greenhouse adaptability of A. potatorum. Methodology: Seven concentrations
of MWCNT (0, 25, 50, 75, 100, 125 & 150 mg L⁻¹) and two of AG₃ (250 & 500 ppm) were chosen at two imbibition
times (24 and 48 h), within a two-phase experimental design. Data were analyzed by ANOVA and Tukey's test (p ≤
0.05). Results: The highest germination percentage was 32% with 125 mg L⁻¹ at 48 h of imbibition, being significantly
higher than the control (p < 0.05). AG₃ inhibited germination with percentages ≤7%, showing phytotoxic effects.
Structural growth of seedlings was 19.6 mm leaf and 5.6 mm root with 25 mg L⁻¹ at 48 h of imbibition. Greenhouse
survival was 100% with 50 and 100 mg L⁻¹ at 48 h of imbibition. Correlation analysis showed a moderate positive
relationship (r = 0.62, p < 0.05) between initial root length and survival. Implications: The use of carbon nanomaterials
(50-125 mg L⁻¹ at 48 h) contributes to increased seed germination, which can be used for field establishment.
Conclusions: MWCNTs improved the germination of A. potatorum seeds, while the use of AG₃ had negative effects
on the seeds under the evaluated conditions.

Triticale malt is an underutilized cereal matrix with high amylolytic potential, which may be exploited for the recovery of plant-
derived enzymes for food and biotechnological applications. This study aimed to obtain α-amylase- and β-amylase-enriched
aqueous extracts from Bicentenario and Siglo XXI triticale malts and to partially purify these enzymes using ethanol/trisodium
citrate aqueous two-phase systems (ATPS). Triticale grains were malted by germination for 5days and dried at 50°C. Aqueous
extraction was performed using ground malt and deionized water at a 1:10 ratio under diferent extraction times (30, 120, and
270min) and temperatures (30°C and 40°C). The selected extracts were lyophilized and subjected to ATPS composed of ethanol
and trisodium citrate. Enzyme activity was determined using the BETAMYL-3 method for β-amylase and the AOAC 2002.01,
AACC 22-02.01, and ICC Standard No. 303 methods for α-amylase. Mild extraction conditions favored enzyme recovery, with the

y fuerte.
